UASt sequences regulate expression of the 'JNK-KTR' kinase translocation reporter of JNK activity (PMID:24949979). 'JNK-KTR' is a fusion protein composed of a JNK docking site from Mmus\Jun, a suboptimal bipartite nuclear localization signal (bNLS), a nuclear export signal (NES) and the Clover fluorescent protein. Both the bNLS and NES have been mutated to contain consensus phosphorylation sites for JNK, such that phosphorylation decreases the activity of the bNLS and increases the activity of the NES. This results in the protein shuttling from the nucleus to the cytoplasm in response to JNK phosphorylation.
