The floxed neo cassette was excised from Gli2tm5(GLI3)Alj, leaving the human GLI3 cDNA, flanked with an upstream FLAG epitope and a downstream mouse Gli2 3' UTR and three tandem repeats of a SV40 polyA signal, positioned within the first coding exon of the endogenous Gli2 locus. Western blot analysis of mutant mice indicated that the full length and the repressor form of the GLI3 protein were present. The level of GLI3 expression from this allele was reported to be greater than from Gli2tm5(GLI3)Alj.
