Description

A targeting vector was designed to insert a PGK-neo and a stop codon in order to splice out exon 5, which encodes the zinc-binding residues in the catalytic domain. Northern blot of mutant calvariae demonstrated a lack of transcript when an exon 5 probe was used and a barely detectable amount of transcript when using a 5' probe. Western blot showed no protein in cultures of mutant calvariae.
