The transgene consists of the human apoE promoter, the human ABCA1 cDNA, the poly(A) signal from the human apoE gene, and the hepatic and macrophage control regions of the apoE/apoC-I locus. Northern blot hybridization analysis and densitometric scanning demonstrated a four-fold increase in encoded mRNA in the liver and a three-fold increase in macrophages. The expressed protein was identified in both mouse liver and macrophage cell extracts by western blotting.
