Nucleotide substitution was used to convert the LSQLL motif in exon 9 encoded by TTAAGCCAACTTCTT to GCAAGCCAAGCTCTT to change the motif to ASQAL. This modification results in an abrogation of protein protein interactions with Nr1h3 and Esr1. A floxed neo cassette was inserted upstream of exon 9 and subsequently removed by cre-mediated recombination. The expression of the modified protein was confirmed by western blot analysis on liver extracts.
