The TgNL1 transgene was designed with a modified regulatory region of the "murine thy1.2 gene" (extending from the promoter to the intron following exon 4, excluding exon 3 and its flanking introns) upstream of a hemagglutinin-tagged mouse neuroligin 1 (Nlgn1 or NL1) cDNA. This transgene was injected into (C57BL/6xCBA)F1 cells. Transgenic founders were bred with C57BL/6 and mice from founder line 6 (TgNL1.6) were found to have approximately two-fold increase in NL1 expression.
