This transgene contains a 40-kDa N-terminal fragment of Mybpc3 (amino acids 1-271, containing the C0 domain, a Pro/Ala-rich region, the C1 domain, and 17 residues of the M domain) under the transcriptional control of a mouse alpha-myosin heavy chain promoter fused to the tetracycline-response element (tetO). An N-terminal c-myc tag encoding the human c-myc peptide was inserted after the initiation methionine codon to differentiate transgenic protein from the endogenous one. Some leakage in the expression of the 40-kDa fragment is seen. Multiple lines were generated, however the pound (#) symbol is used because line number is not specified.
