Description

This transgene consists of a 36.8-kb SwaI fragment of the BAC clone RP23-551L11 (from 129S6/SvEvTac) that contains the complete Wnk4 transcription unit plus 9 kb of 5' flanking and 11.5 kb of 3' flanking sequence. Two substitutions were introduced: one that creates the pseudohypoaldosteronism type II (PHAII) amino acid substitution Q562E and a synonymous substitution at codon 563, creating a new Bsp1286I cleavage site. Fluorescence in situ hybridization showed a single site of insertion. Transgene expression was detected in heart, kidney, testis, brain, and lung but not in liver or spleen. RT-PCR showed that total amount of mRNA is increased by about 2-fold. The pound symbol (#) is used when line is not specified and/or lines are pooled.
